Ph D candidate for the project ''Characterization of fusion inhibitory peptides against measles virus''
Job description
Respiratory viruses are omnipresent, affecting global health and causing economic losses. Development of novel intervention strategies to combat these viruses is essential. The most recent example underlining this importance is the recent COVID-19 pandemic.Despite the availability of safe and effective measles vaccines, the global incidence of measles is on the rise. Vaccine hesitancy is an important underlying problem, but there are other factors that contribute to the resurgence of measles. Since measles vaccines contain live-attenuated measles virus, patients with severe immunodeficiencies cannot be vaccinated. In addition, there is a so-called immunity gap: a period during which infants (6-14 months) have lost their maternal antibodies but are not yet eligible for vaccination and therefore susceptible to measles.
Together with collaborators in the US, we are evaluating specific antiviral peptides that can interfere with the membrane fusion between virus and target cell, thereby preventing virus infection. In initial proof-of-concept studies we have shown that these fusion inhibitory peptides can prevent measles virus infection in vitro.
In this project, the Ph D candidate will help with the design of peptides and will design and perform experiments to demonstrate in vivo efficacy of these peptides. Additionally, the Ph D candidate will work on improvements of the peptides with regards to solubility, toxicity, stability, and biodistribution.
In these studies, the candidate will use state-of-the-art techniques, including (primary) cell culture, flow cytometry, comprehensive data-set analysis, multicolor confocal microscopy, immunological assays, virus culture, and animal experiments in a BSL-2 and -3 environment.
Work environment
A healthy population and excellence in healthcare through research and education. This is what Erasmus MC stands for. Conducting groundbreaking work, pushing boundaries and leading the way. In research, education, and healthcare. We are practical people with a high level of expertise, working hard to improve and renew the healthcare of today and the public health of tomorrow. The Department of Viroscience at Erasmus MC employs over 180 FTE and exists of the unit Clinical Virology and the unit Research. Within the Clinical Virology unit, viral diagnostics are conducted for patient care within the Netherlands and internationally. The unit focuses on continuous development of new and improved diagnostic methods, as well as patient-centred topics such as antiviral resistance, viral pathogenesis, and the effectiveness of antiviral therapy. The unit Research harbors different disciplines and runs an extensive research program on the epidemiology, pathogenesis, prevention, and treatment of infections by human and animal viruses. The clinical studies are performed in close collaboration between the two units.
